Changes in vasodilation via protease-activated receptor-2 with ageing differ between animal models of metabolic syndrome

Abstract: The worldwide prevalence of metabolic syndrome is increasing and becoming a global health problem. Metabolic syndrome is a chronic low-grade systemic inflammation condition as evidenced by the release of inflammatory cytokines from adipose tissue that is associated with development of obesity, insulin resistance and cardiovascular diseases, including endothelial dysfunction. Some of these cytokines induce protease-activated receptor-2 (PAR2) expression which is not only associated with chronic inflammation and tissue injury, but also regulation of vascular tone. In SHRSP.Z-Leprfa/IzmDmcr rats (SHRSP.ZF) with metabolic syndrome, PAR2-mediated endothelium-dependent vasodilation is preserved in aortas at 20 weeks of age (wks), but then decreases at 30 wks. Since oxidative stress was also higher at 30 wks and correlated with the degree of impaired PAR2-mediated vasodilation, we proposed that increased oxidative stress modulates PAR2 vascular function.
