Gene-Editing Technologies and Applications for Molecular Imaging
Year: 2021
Type: book-chapter
Abstract: Gene-editing technologies encompass a variety of methods to alter structure and composition of DNA and RNA, including changing specific bases, deleting bases or segments of DNA, and inserting new DNA sequences in defined reading frames. Recent advances in CRISPR-Cas tools and methods now make gene editing readily accessible to labs involved in molecular imaging. Gene-editing tools open potentially transformative new approaches to advance molecular imaging through applications including introducing imaging reporters under regulation of endogenous promoters, generating fusion proteins expressed at physiologic levels, and tracking dynamic changes in chromatin structure and positions of specific genes. This chapter reviews basic mechanisms underlying gene editing, technologies to achieve gene editing in cultured cells and animal models, and selected applications in molecular imaging. Methods for facile editing of genes continue to open exciting new opportunities for technology development and discovery in molecular imaging.
