Abstract: The development of new assays to process high number of samples in a short period of time is a clear nowadays. This paper presents an alternative test to standard OECD protocol for the Algal Growth Inhibition Test, using Chlorella vulgaris, an unicellular alga. The assay organism has been grown like a cellular culture under assay conditions which allowed a quick cellular growth, ten times higher than initial culture. A linear relationship between Absorbance and time was obtained for an Absorbance range between 0.4 and 1.25 in 48 hours. 3,4-dichloroaniline IC50 was calculated using this alternative method and results were compared with standardized test results. The IC50 values were 3.01 ppm at 72 hours and 3.75 ppm at 48 hours for the standard and the microplate test, respectively. The results suggest the capability of the microplate test, specifically to manage low sample volumes, a critical requirement for Toxicity Identification Evaluation (TIE) protocols.
