Induction of 11 beta-hydroxylase by corticotropin in primary cultures of bovine adrenocortical cells.

Abstract: Primary cultures of bovine adrenocortical cells were used to evaluate the trophic effect of adrenocorticotropin (ACTH) on 11/3-hydroxylase.Incubation with ACTH (lo-' M) for 36 h resulted in a marked increase in the synthesis of cytochrome P-4501, in both radiolabeled cells and in a cell-free translation system directed by adrenocortical cell RNA, an effect paralleled by an increase in the synthesis of cytochrome P-450.,,.Mitochondrial cytochrome P-450 content and the magnitude of the 11-deoxycorticosterone-induced type I absorbance change were increased 2-fold by ACTH.In addition, mitochondrial 11/3-hydroxylase activity as well as the ability of adrenocortical cells to convert exogenous 11-deoxycorticosterone to corticosterone were increased 2.5-fold 36 h after the initiation of ACTH treatment.By 72 h, however, the actions of ACTH on the synthesis of cytochrome P-45Ol1p and on the conversion of 11-deoxycorticosterone to corticosterone by adrenocortical cells declined 5 0 8 , while ll/?-hydroxyIase activity assayed in isolated mitochondria declined to levels 25% of control values. In contrast to 11,!?-hydroxylase synthesis and activity, cortisol production was maximal after 12 h of incubation with ACTH and declined steadily thereafter. These results are indicative that ACTH has long term effects to promote the synthesis and activity of ll~-hydroxylase and together with results of other studies carried out in this labora-
