Homologies Between the Genetic Etiology and Pathogenesis of the Synucleinopathies and Tauopathies
Year: 2003
Type: book-chapter
Abstract: Two major classes of neurodegenerative disease are those in which tangles occur and those in which Lewy bodies occur. Tangles are made up largely of the protein tau (1) and Lewy bodies are made up largely of the protein α-synuclein (2). Recent genetic and pathological data show many relationships and similarities between these two classes of disease. Parkinson’s disease (PD), lewy body dementia (LBD), progressive supranuclear palsy (PSP), and corticobasal degeneration (CBD) are diseases in which there is intracellular inclusions of synuclein containing Lewy bodies (PD and LBD) or tangles containing tau protein (PSP and CBD). These same lesions can occur in Alzheimer’s disease (AD) and prion disease (PrD) when they are secondary to the initiating lesions (amyloid β [Aβ] in AD and prion proteins Sc in PrD).
