Abstract: This protocol describes the process/settings to sort single nuclei into 96-well plates by fluorescence-activated cell sorting (FACS) for Smart-seq2 using the MoFlo Astrios EQ cell sorter (Beckman Coulter). This protocol was applied to frozen pediatric glioma samples for the Human Tumor Atlas Pilot Project (HTAPP). [Note: It does not include instructions on how to operate the cell sorter Beckman Coulter MoFlo Astrios EQ.]
