A Role for Dopamine-Mediated Learning in the Pathophysiology and Treatment of Parkinson’s DiseaseBeeler et al.Cell ReportsDecember 13, 2012In BriefAlthough it has been demonstrated that dopamine denervation induces abnormalities in corticostriatal plasticity, the role this plays in Parkinson’s disease remains speculative. Beeler and colleagues show that dopamine blockade induces a D2-mediated aberrant learning that impairs future motor performance, even when dopamine is restored, suggesting abnormal corticostriatal plasticity is structurally encoding inappropriate inhibitory learning. Such aberrant learning may contribute significantly to the motor symptoms observed in Parkinson’s disease and represent an important target for the development of new therapeutics.
