Perm1 Protects the Heart From Pressure Overload–Induced Dysfunction by Promoting Oxidative Metabolism

Year: 2023
Type: article
Abstract: Defects in ATP production lead to cardiac dysfunction and heart failure (HF), while maintaining appropriate energy production preserves normal heart function and restoring reduced mitochondrial (Mito) function mitigates HF in human and animal models. Therefore, modulating cardiac energy production has therapeutic potential 1 .Previously we showed a novel protein Perm1 (PGC-1 and ERR induced regulator, muscle 1) promotes Mito biogenesis in cardiomyocytes (CMs) 2 .The role of myocardial Perm1 remains poorly understood, so mice were produced to increase Perm1 specifically in CMs.CAG-STOP-Perm1 mice were constructed (Fig Ai ), which when interbred with αMyosin Heavy Chain (MHC)-nuclear Cre mice, produced CM-specific overexpression of Perm1.UC San Diego IACUC approved all animal protocols. These mice are termed Perm1cTG.
