Abstract: Supplemental Figure 6. (A & B) OCI-AML3 cells were transduced with CTRL shRNA, PKCε shRNA_1 or PKCε shRNA_2 and 24 hours later cells from each condition were treated with 100µM NAC (A) or 50µM glutathione (B), every 12 hours until the end of the experiment. Four days after treatment, cells from each condition were assessed for Annexin V staining by flow cytometry (****p<0.0001). OCI-AML3 cells were also treated with 20 µM Menadione with and without NAC of glutathione as a positive control for anti-oxidant activity. Data are represented as mean {plus minus} SD of three technical replicates. (C) Mouse MLL-AF9 leukemia cells were transduced with CTRL shRNA or Pkcε shRNA. Forty-eight hours after transduction, cells were treated with 25M BHA or DMSO. Forty-eight hours after BHA treatment, cells from each condition were assessed for Annexin V staining by flow cytometry (*p{less than or equal to}0.05). Data are represented as mean {plus minus} SD of three technical replicates. (D) OCI-AML3 cells stably transduced with retroviruses co-expressing SOD2-IRES-Catalase (SOD2-Catalase) and GFP or control vector (CTRL) were transduced with CTRL shRNA, PKCε shRNA_1 or PKCε shRNA_2. Four days after transduction, cells were analyzed for mitochondrial superoxides production. Data are represented as mean {plus minus} SD of three independent experiments for each panel. (*p{less than or equal to}0.05;**p{less than or equal to}0.01).
