Abstract: Sequence alignment to reference genome reveals an acquired C6097G>A (D2033N) mutation in ROS1 after treatment with crizotinib. The MSK-IMPACT (Integrated Mutational Profiling of Actionable Targets, Illumina HiSeq 2500, 341-gene panel) platform was used for broad, hybrid-capture next-generation sequencing of (A) pre- and (B) post-crizotinib tumor tissue. Data is displayed via the Integrated Genomics Viewer (IGV). A novel acquired ROS1 D2033N mutation (c.6097G>A) was detected in the post-crizotinib treatment tumor sample but was not detected before treatment.
