Abstract: Supplementary Table 1. Patient Demographic Characteristics. *2 patients with metachronous tumors, 1 poor and 1 moderate; **One patient with both pre- and posttreatment IHC and MSI testing. Supplementary Table 2. Tumor characteristics and results of the IHC and MSI analysis of those cases exhibiting loss of MSH6 after neoadjuvant treatment. Abbreviations: yp, pathologic stage after neoadjuvant treatment; NT, not tested; pre-tx, pre-treatment; post-tx, post-treatment; MSS, microsatellite stable; MSI-L, microsatellite low. Supplementary Table 3. MSI and IHC Analysis of in vivo pre- and post-treatment data. Abbreviations: yp, pathologic stage after neoadjuvant treatment; pre-tx, pre-treatment; post-tx, post-treatment; MSS, microsatellite stable; MSI-L, microsatellite low. Supplementary Table 4. PDX Patient Demographic Characteristics (n = 5) Supplementary Figure 1. Confirmation of MLH1 loss in the cell line HCT116 was performed via Sanger sequencing (A) and Western Blot (B) Supplementary Figure 2. In vitro treatment method. MMR deficient (HCT116) and proficient (HCT116+Ch3) isogenic cell lines were exposed to treatment every 48 hours for 3 cycles. On Day 1, 500,000 cells/10 mm plate were seeded and allowed to grow exponentially for 24-48 hours. Cells were then treated with radiation or drug. After 48hrs, the cells were allowed to grow in drug-free medium until confluency. Cells were split and allowed to grow again in drug-free medium for 48 hours before being treated with the next round of treatment. This process was repeated for 3 cycles. After 48 hours of the 3rd treatment, the slides were fixed with 4% paraformaldehyde.
