Abstract: Fig. S2. (A) Metaphase spread of the cell line AG09391 hybridized with the NFIB(SG)/MTFR2(SR) FISH fusion probe set. The MTFR2 probe recognized homology with the chromosome 6q terminal band, compatible with mapping at 6q23.3; the NFIB probe recognized homology with the chromosome 9p terminal band, compatible with mapping at 9p22.3. Normal CUSG004 patient tissue hybridized with the NFIB(SG)/MTFR2(SR) FISH fusion probe set, showing negative pattern. CUSG004 patient tumor tissue (F0) and PDX tumor tissue (F1, F2, F5) hybridized with the NFIB(SG)/MTFR2(SR) FISH fusion probe set, with patient tumor and PDX tissue showing positive pattern. (B) CUSG005 patient (F0), CUSG005 PDX (F1), and CUSG006 PDX (F2) tumor tissue hybridized with the NFIB(SG)/MTFR2(SR) FISH fusion probe set, with CUSG005 tissue showing positive pattern and CUSG006 tissue a negative pattern. Our FISH probes also detected the MYB-NFIB fusion in the CUSG005 tumor and was likely due to the close proximity (<1Mbp) of MTFR2 and MYB on the q arm of chromosome 6 Arrows indicate fused signals.
