Abstract: PDF file - 247K, The effect of the silencing of c-MET protein by siRNA on MPM cell lines. siRNAs targeting c-MET were designed with siDirect online software system (http://www.rnai.co.jp/e_index.html). siRNA sequences targeting two different regions of c-MET are as follows (sense and antisense, respectively): si-c-MET-1 (5'-CAUUGCAUUCGAUAUCAGUGA-3' and 5'-ACUGAUAUCGAAUGCAAUGGA-3') and si-c-Met-2 (5'-CAUGUCAACAUCGCUCUAAUU-3' and 5'-UUAGAGCGAUGUUGACAUGCC-3'). Cells were transfected with 20 nM of siRNA using LipofectamineTM RNAiMAX (Invitrogen) according to the manufacturer's protocol. Control cells were treated with RNAiMAX alone or with scramble. Cell viability after siRNA introduction was determined by a MTS assay.
