Abstract: The ROS1D2033N mutant retains transforming capacity but is resistant to crizotinib. A. IL-3 withdrawal assay for Ba/F3 cells retrovirally transduced with native CD74-ROS1 or CD74-ROS1D2033N. Total viable cell number was determined by counting cells on days 2, 4, and 6 after removal of IL-3 from the culture medium. B. Apoptosis induction in Ba/F3 CD74-ROS1 and CD74-ROS1D2033N cells after 72 h of inhibitor treatment at the indicated concentrations. Percentage of cells that were Annexin V-positive after inhibitor treatment is indicated as % Apoptotic. 2,000 cells were counted per condition.
