Abstract: Figure S5. Magnitude and breadth of T-cell response to HPV-16 E7/E6 DNA vaccine. The E7/E6 DNA vaccine (VB1025) was obtained from Agnete Fredriksen and Ole Brekke (Vaccibody, Oslo, Norway). It encodes a HPV-16 E7/E6 fusion protein, which for safety reasons, has mutations at E7 position 24 (Cys to Gly) and 26 (Glu to Gly), and E6 position 63 (Cys to Arg) and 106 (Cys to Arg). C57BL/6J mice were immunized with a total of 50 µg of empty vector or VB1025 plasmid DNA in normal saline injected into both quadriceps. After 12 days, the number of epitope-specific T-cells induced was assayed directly ex vivo by IFN-γ ELISPOT. Overlapping 15-mer peptide pools (5 per pool) spanning the E7/E6 fusion protein or the nominal CD8+ T-cell epitopes RAHYNIVTF (derived from E7 and represented in pool 3) and EVYDFAARR (derived from E6 and represented in pool 8) were used to determine the breadth of the response. Each assay was performed in triplicate, and the experimental values were expressed as the mean net spots/106 splenocytes {plus minus} SD of the mean for each peptide pool tested. The results shown are representative of two separate experiments, with each column representing an individual mouse.
