Abstract: PDF - 95K, Supplementary Figure 3. Morphometric analysis of CTTN distribution. Intensity of signal for IF staining for CTTN and actin was calculated based on 3D reconstracted confocal projections of the whole cell along the line drawn across the cell length starting from morphologically distinct lamellipodia edge-point A on cell diagram (also on Fig.3A), through the nucleus towards the rear end of the cell- point B on cell diagram (also on Fig.3A). Intensity of staining was counted using ImageJ for each pixel along the line as a Sum of Max Grey value in each of 10 confocal sections taken at 0.2!m steps comprasing the Z-stack for each cell. MDA-MB-231 cells nucleofected with either siCon or siNEDD9 (10 cells per group) were evaluated. Length of each cell was taken as 100% and distribution of CTTN and actin in relation to percent of the cell length was calculated.
