Abstract: PDF file - 627K, (A) Effects of 200 nM SAHA on mRNA expressions of DSB repair-related genes in response to IR. MM cells were treated with SAHA for 16 hours, followed by 6 Gy IR. Total RNA were prepared 3 and 24 hours later, and subjected to RPA assay with custom-designed probe sets for DNA repair protein profile. (B). Time dependent effects of SAHA on mRNA expressions. U266B1 cells were exposed to 200 nM SAHA for indicated time periods, and total RNA prepared were subjected to RPA assay. (C) Dose-dependent effects of SAHA on mRNA and protein levels of RAD51. U266B1 cells were exposed to vairous concentrations of SAHA for 16 hours, and total RNA prepared were subjected to RPA assay. Western blot shows higher concentrations of SAHA reduce RAD51 protein level in U266B1 cells
