Abstract: (A) Positive responses to overlapping peptide pools by ELISPOT. (B) Positive responses to individual peptide by ELISPOT. Intra-assay variability was based on 8 peptides of patients 6, 53, and 110. Each of which was run in duplicate on the same plate. The within-plate percent coefficient of variation for each peptide was calculated by dividing the standard deviation of each replicate pair by its mean and multiplying by 100. The average of the peptide coefficients of variation is reported as the intra-assay coefficient of variation. The intra-assay variability was 10.5%. The inter-assay variability was based on four different peptides (ATP 13, 14, 23, and 32) that were run in duplicate on two different plates to monitor plate-to-plate variation. Plate means were calculated for each peptide. The pairs of plate means were used to calculate a mean, standard deviation, and percent coefficient of variability for each peptide. The inter-assay variability reported is the average percent coefficient of variability of the four peptides. The inter-assay variability was 25.6%.
