Abstract: Supplemental Figure 1. A. Survival of GBM9 tumor-bearing, untreated or TMZ-treated mice was recorded and represented in a Kaplan-Meier plot. *, p=0.0253 (untreated vs. TMZ treated). B. Primary (483 and 484, in black) and recurrent (419 and 481, in green) cultures were treated with 10 Gy of IR, and cell cycle distribution was assessed by single-parameter flow cytometry after propidium iodide staining for DNA content (x-axis). Please note that all lines are capable of G2 arrest in response to ionizing radiation. C. Primary and recurrent cultures were either mock-irradiated (M) or treated with 10 Gy of ionizing radiation (IR), and harvested after 30 minutes. Whole cell lysates of irradiated cultures were probed for CHK1 and CHK2 phosphorylation by Western blotting with phospho-specific antibodies. Please note activation of CHK1 and CHK2 in both primary and recurrent cultures.
