Abstract: (A) Peak area for each peptide for a protein (exemplified by MAPK3 - ALLDLLR) as analyzed by liquid chromatography-multiple reaction monitoring (LC-MRM). Peak area for each protein (representative peptide) was ranked from 1-15 based on the levels with 1 being the highest level and 15 being the lowest level (B) Heatmap indicating the cell line ranking for each kinase to assess the relative expression level of each kinase in the 15 cell lines. Cell lines were ranked from 1 to 15. (C) WEE1, pY15-CDK1, and total CDK1 levels in 21 untreated log growing lung cancer cell lines. (D) Dose-response curves of three lung cancer cell lines with varying expression of WEE1. The cell lines expressing WEE1 had significantly lower IC50s than those with no expression of WEE1 (H322: 0.45 ï�M, H1648: 1.23 ï�M, H1395: >10 ï�M, %RLU - Relative light units measuring ATP).
