Abstract: Ex-vivo depletion of HA from KPC tumor sections with hyaluronidase and PEGPH20. To confirm specificity of HA staining, 14-day orthotopic KPC tumors were extracted, flash frozen in OCT medium, sectioned, and fixed onto Colorfrost slides (Fisher Scientific) with acid formalin. Sections were then incubated with 1 mg/ml bovine testis hyaluronidase (Sigma) , PEGPH20, or left untreated for 1 hour in a humidified 37degreeC incubator. Sections were then incubated with biotinylated HA binding protein (HABP, 5ug/ml) overnight at 4degreeC followed by treatment using an avidin-biotin peroxidase kit (ABC kit, Vectastain).
