Abstract: Supplemental Table 4. NK cells were negatively enriched from pooled spleens of 3 WT and 3 Cd47-/- littermate mice and cultured with/out thrombospondin-1 (2nM) for 24 hours in RPMI (without FCS) + IL-15 (2.5 ng/ml). Golgi-stop was added at last 4 hours of culture. FcRs were blocked with rat and rabbit serum and cell surface CD47 and NK1.1 were stained with Aqua Live/Dead. Cells were then fixed, permeabilized and i.c. stained for Ki-67, IL-2, IFNγ, TNFα and GzmB. Values indicate the MFI of indicated proteins.
