Abstract: Despite its universality, the molecular mechanisms underlying the aging process remain poorly understood. Liu et al (Cell 2023;186:287-304.e26; https://doi.org/10.1016/j.cell.2022.12.017) uncovered a positive feedback loop between the activation of dormant endogenous retroviruses and aging. More specifically, cellular senescence induced the reactivation and accumulation of the endogenous retrovirus HERVK. This in turn led to the production and extracellular release of retrovirus-like particles (RVLPs) that induced cellular senescence in neighboring young cells in a paracrine fashion.
