Abstract: PDF - 1449KB, PAI-1 expression in T24-PAI-1KD-19, T24-PAI-1KD-22, UM-UC-14-PAI-1KD-4, UM-UC-14-PAI-1KD-17, HeLa-PAI-1OE-12 and HeLa-PAI-1OE-18 clones was confirmed by immunoblotting (A), quantitative PCR (B), and secreted protein by ELISA (C) and compared to control. D, Protein lysates from T24, UM-UC-14, UROtsa and HeLa cells treated with or without tiplaxtinin (0-50 microM) and from the above clones were electrophoresed on 10% SDS-polyacrylamide gels containing plasminogen and uPA under non-reducing conditions followed by staining with Coomassie blue. PAI-1 activity was indicated as lytic zones of plasmin generation. Specifically in our in vitro zymogen activity assay, we confirmed in a dose dependent manner that tiplaxtinin stimulates the conversion of zymogen to mature enzyme (plasmin) resulting in an increase in enzymatic activity. Similar, clones in which PAI-1 was silence noted conversion of zymogen to mature enzyme resulting in an increase in enzymatic activity. *, p < 0.05.
