Abstract: (A) WEE1 In vitro kinase assays. Purified GST-tagged WEE1 and active GST-CDK1 were incubated in the presence or absence of recombinant GST-PAXIP1 tBRCT C2. Arrows indicate the bands for auto-phosphorylated WEE1 and phosphorylated CDK1. Note the absence of a phosphorylated band corresponding to the expected size of the recombinant PAXIP1 tBRCT C2. (B) Experimental design timeline for cell cycle assays. (C-F) H322 cells transfected with: (C) empty vector; a vector expressing PAXIP1 (D); a vector containing a scrambled control shRNA (E) or PAXIP1 shRNA (F) were mock-treated or pre-treated with AZD1775 for 1 h prior to receiving 6 Gy of IR. Cells were incubated for 4 hours and cell cycle analysis was performed to test the mitotic index (percentage of phospho-histone H3 positive cells) at the early G2/M checkpoint post IR.
