Abstract: Supplementary Figure 4 A. Splenocytes from E7-TCRβ transgenic mice were stained with a tetramer for HPV E749-57 (RAHYNIVTF) or a non-specific control tetramer. Cells were gated on CD3+ CD8+. B. Splenocytes from E7-TCRβ mice were stimulated for 7-9 days using HPV E749-57 (RAHYNIVTF) peptide and rhIL-2 (30u/ml). CD8+ T cells were then purfied by magnetic bead purification and used in an in vitro CTL assay using irradiated, labeled or CFSE labeled cells as targets. Ratios are pulsed E7 TCRβ effectors : unlabled control target cells : CFSE-labeled target cells. Wildtype, unlabeled EL4 cells served as negative control targets. C. CD8+ T cells were purified from E7-TCRβ splenocytes using negative magnetic bead selection and transferred into TC-1 tumor bearing mice in the indicated numbers on day 7 after tumor injection.
