Abstract: Gating strategy for three-color flow cytometry analysis of splenic MDSCs. Splenocytes were simultaneously stained with anti-CD11b-FITC, anti-Gr1-PE and anti-Ly6C-APC antibodies or the corresponding isotype control antibodies and analyzed by flow cytometry as follows. Gate 1: single cells were gated and cell aggregates were excluded. Gate 2: using side-scatter (SSC) and forward-scatter (FSC), granular and non-granular (mononuclear) cells were gated. Gate 3: CD11b+ granular (A) and mononuclear (B) cells were gated. Gate 4: Gr1 and Ly6 expression on CD11b+ granular (A) and mononuclear (B) cells was examined.
