Abstract: Treatment-emergent T790M by baseline EGFR-activating mutation subgroup and cumulative frequencies over time. The population consisted of RELAY patients who had an available liquid biopsy sample at baseline with a valid next-generation sequencing (NGS) result with any gene alteration detected and/or at least a valid NGS result with the post-study treatment discontinuation liquid biopsy sample that was also positive for the original EGFR-activating mutation (ex19del or ex21L858R). Analysis population includes patients (N=119) with disease progression and detectable EGFR ex19del or ex21L858R at 30-day follow-up to ensure inclusion of patients shedding tumor DNA at progression. There are two time points shown in the plots below: baseline and 30-day follow-up. Each set of bars (other than baseline) is a subset of the 30-day follow-up samples based on how many cycles of treatment the patient completed before progression. Panels (A) and (B) show post-progression T790M frequencies at the 30-day follow-up visit using NGS; (C) and (D) show the cumulative incidence of post-progression T790M as a function of the maximum number of treatment cycles received prior to disease progression.
