Abstract: Antigen-loaded, biodegradable PLG nanoparticles have demonstrated tolerance induction in models of autoimmune disease. Here we report on the effect of these particles on transgenic CD8+ T cells. Wild-type mice received transgenic cells, were treated with nanoparticles, and then had their spleens and/or pancreases removed for flow cytometric analysis. Relative to controls, mice treated with these particles exhibited a larger percentage of transgenic CD8+ T cells in spleens, suggesting that a sequestration of autoreactive T cells prevents their trafficking to inflamed tissues. Furthermore, nanoparticle treatment led to a higher percentage of cells with surface markers indicative of a regulatory phenotype. These results imply that altered trafficking and an expansion of regulatory T cells are two mechanisms by which PLG nanoparticles induce tolerance in CD8+ T cell - mediated autoimmune disease.
