Abstract: Lockhart et al (J Exp Med 2023;220:e20221816; https://doi.org/10.1084/jem.20221816) identified an important tolerogenic role for dietary protein in C57BL/6J mice. Food proteins ingested during homeostasis induced polyclonal antigen-specific expansion of CD4+ T cells in the intestinal epithelium with enriched cytotoxic gene expression. Notably, this process was enhanced by commensal bacteria. Furthermore, active tolerance was associated with influx of antigen-experienced regulatory T cells whereas polyclonal expansion of proinflammatory CD4+ T was observed in the contexts of food allergy.
