Abstract: Introduction: In the past couple of decades, rapid growth has been observed in the field of traditional medicine. It is estimated that 80% of the population of developing countries relies on traditional medicines for their primary health-care needs. Henceforth, the traditional systems of medicine are required to develop essential techniques of standardization for their universal acceptance. Amrtottara Kwatha Churna (AKC) is a polyherbal Ayurveda formulation being used frequently across the country. However, its analytical profile is not available as on date. Aim: The aim of this study was to develop preliminary analytical profile for AKC and determine catechin and berberine content using high-performance thin-layer chromatography (HPTLC). Materials and Methods: The individual ingredients of AKC that fulfill the quality standards were procured from local market, and AKC was prepared following standard operative procedures as mentioned in the classical literature. The finished product was evaluated through macroscopical, microscopical, powder flow properties, extractive values, physicochemical characters, heavy metal content, and HPTLC fingerprinting. Results: The preliminary phytochemical test revealed the presence of several bioactive constituents such as polyphenolics (tannins, phenolics, and flavonoids), saponins, steroids, proteins, amino acids, and carbohydrates in its hydroalcoholic extract, revealing its significant therapeutic action. The quantity of catechin and berberine present in AKC methanolic extract was found to be 9.52% and 7.1% (w/w), respectively, which may serve as a chemical marker for future studies on the formulation. Conclusion: This study encompasses an initial quality control assessment of various measures and HPTLC fingerprinting of AKC. The preliminary analytical profile may serve as a significant tool to ensure the authenticity and to ascertain the quality of the formulation.
