Quantitation of Ca2+ Release Channel mRNA in Pressure-overload Induced Hypertrophic Rat Heart by Competitive Reverse Transcription and Competitive Polymerase Chain Reaction

Type: article
Abstract: The density of calcium release channels (CRC) in pressure-overload induced hypertrophic rat hearts is significantly lower than that in sham-operated control animals (Kim et al., 1994). The present study was undertaken to assess the expression levels of CRC in sham-operated and hypertrophied animals at the transcriptional level by competitive reverse transcription and competitive polymerase chain reaction (cRT-cPCR). Severe left ventricular hypertrophy (>70%) was induced by banding the abdominal aorta. To estimate the copy number of CRC mRNA by cRT-cPCR, CRC mRNA templates of sham-operated and hypertrophied left ventricles were amplified with varying amounts of CRC competitor RNA. As a control, glyceraldehyde phosphate dehydrogenase (GAPDH) mRNA levels were determined similarly. Our results show that CRC copy number was significantly decreased by pressure-overload induced hypertrophy (1.15 ±0.22 × 1010 vs. 2.03 ±0.20 × 1010/μg mRNA, p≤0.05), whereas GAPDH copy numbers were similar in sham-operated and hypertrophied animals. These results suggest that CRC expression is specifically down-regulated at the transcriptional level during the development of left ventricular hypertrophy by pressure-overload.
