Joint modeling of genomic and exposomic data reveals a substantial GxE component in type 2 diabetes etiology

Abstract: Understanding the relative contribution of genetic and environmental factors and their interactions is important in mapping complex diseases. We integrated genomic data with unprecedented questionnaire-based exposure data from the Personalized Environment and Genes Study (PEGS) to dissect the etiology of type 2 diabetes and two exemplar traits, namely height and BMI. We estimated the phenotypic variance for type 2 diabetes contributed by the genome, exposome, and their interactions (50.96%, 2.80%, and 18.36%, respectively). The results demonstrate the utility of incorporating exposomic data and considering gene-environment interactions when seeking insights into the causes of common, complex diseases.
