Modeling challenges of hepatitis D virus kinetics during bulevirtide-based therapy

Year: 2024
Type: article
Abstract: We read with interest the study by El Messaoudi et al.1 analyzing and mathematically modeling serum HDV RNA and biochemical (ALT) kinetics during therapy with the entry inhibitor bulevirtide (BLV) alone or in combination with pegylated interferon (Peg-IFN). The authors analyzed an impressive amount of kinetic data from 183 treated patients (114 BLV; 69 BLV+Peg-IFN) who participated in two French multicenter cohort studies. They used a mathematical model to estimate BLV efficacy in blocking infection (parameter η) and viral and host kinetic parameters, such as the loss rate of HDV-infected cells (parameter δ), to predict the optimal duration of BLV-based treatment and rate of cure.
