Advances in gluten detection: A rapid colorimetric approach using core-satellite magnetic particles

Year: 2024
Type: article
Abstract: A colorimetric immunosensor for the rapid and sensitive detection of gluten in flour-based food products is described. This immunosensor exploits core-satellite magnetic particles (CSMPs) – magnetic cores with gold nanoparticles (AuNPs) on their surfaces – that have been functionalized with anti-gliadin antibodies, which bind to the gold surfaces. The study demonstrates that an appropriate concentration of surfactant Tween-20 can induce the spontaneous formation of stable clusters of CSMPs in the dispersion. These clusters consist of weakly interacting CSMPs, and their fragmentation, induced by the presence of gliadin, leads to a color change. Gliadin extraction using a non-toxic ethanol-water mixture is sufficient to give rise a measurable color change in the presence of gluten contamination at levels of less than 10 ppm.
