Abstract: LHMM24001 EFFECT OF GIGANTOL ON THE TNF-Α/NF-ΚB PATHWAY IN DIABETIC RETINOPATHY Yahan Luoa,b, Guifu Lib, Chunxia Lib aShanghai University of Traditional Chinese Medicine, Shanghai 201203, China,bDepartment of Ophthalmology, Shanghai TCM-Integrated Hospital, Shanghai University of TCM, Shanghai, 200082, China. Background: The purpose of this study was to investigate the effect of gigantol on retinal cell injury induced by high glucose (HG) culture in rhesus monkey choroidal-retinal endothelial (RF/6A) cells and in mouse models of diabetes in vitro and in vivo. Subjects and Methods: We firstly detected the effect of gigantol on proliferation of RF/6A cells with the CCK-8 kit. Secondly, we used an NF-κB reporter assay to determine whether gigantol has anti-inflammatory activity against the NF-κB gene. Thirdly, RF/6A cells were treated with different concentrations of gigantol (5, 10, or 25 μmol/ L) and cultured under HG conditions (30 mmol/ L) or TNF-α (5 ng/ ml) for 48 h. Afterwards, we observed changes in the TNF-α/NF-κB pathway protein in RF/6A cells, and detected the effect of gigantol on TNF-α/NF-κB pathway related gene expression in RF/6A cells. In a mouse model of STZ-induced (55 mg/kg) diabetes, the protein levels of TNF-α and interleukin-6 (IL-6) in the serum were analyzed using ELISA. The expression of NF-κB in the retinal tissue of each group was detected by immunohistochemistry and the protein and related gene expression of TNF-α/NF-κB pathway observed in mouse retina. Results: Our findings show that gigantol can inhibit NF-κB transactivity. Gigantol inhibited high glucose and TNF-α induced NF-κB expression in RF/6A cells. The mRNA and protein levels of inflammatory cytokines downstream of NF-κB were also reduced. In a mouse model of STZ-induced diabetes, gigantol reduced serum TNF-α and IL-6 expression levels and prevented inflammatory damage to the retina. Conclusions: Our data suggest that gigantol may exert anti-inflammatory effects by inhibiting the NF-κB signalling pathway in diabetic retinopathy. Our findings are valuable for understanding the mechanism of action of gigantol and may be useful for drug development or food supplement research for diabetic retinopathy. LHMM24002 OBSERVATION OF THE THERAPEUTIC EFFECT OF POSTERIOR SCLERAL REINFORCEMENT IN THE TREATMENT OF PATHOLOGICAL MYOPIA IN DIFFERENT AGE GROUPS Enguang Dinga, Enliang Dinga, Dan Menga, Yongxia Zhanga, Hongyu Sua, Yongli Lia, Yuxuan Haob, Lijuan Suna aHanDan AiYan Ophthalmology Hospital, Handan Hebei 056008, China,bClinical Medicine, Hebei Medical University, Shijiazhuang, Hebei 050000, China. Background: To compare the efficacy and safety of posterior scleral reinforcement (PSR) in the treatment of pathological myopia in different age groups. Subjects and Methods: 27 patients (31 eyes) with pathological myopia who underwent PSR treatment were selected and divided into two groups based on age. Patients aged>18 years old were classified as Group A, and those aged ≤ 18 years old were classified as Group B. The follow-up period shall not be less than 6 months, and the best corrected visual acuity (BCVA), refractive index, axial length, and complications of the two groups of patients shall be observed before surgery, 1 day after surgery, 3 meters after surgery, and the last follow-up. Results: The follow-up time for Group A was (8.76±3.11) months, and for Group B was (9.00±3.08) months. There was no significant difference in BCVA between the two groups of patients at postoperative 1 day, 3 meters, and the last follow-up (P>0.05), and there was no difference in BCVA changes between the two groups during each follow-up period; The refractive index of the two groups of patients decreased compared to preoperative one day and three months after surgery (P<0.05). There was no significant difference in refractive index between the last follow-up and preoperative, and there was no difference in refractive index changes between the two groups of patients during each follow-up period; The axial length of the eyes of the two groups of patients was shortened compared to before surgery at 1 day, 3 months, and the last follow-up, and the difference was statistically significant. One day after surgery, three months after surgery, and the last follow-up, Group A had more axial reduction than Group B, and the difference was statistically significant. On the first day after surgery, 7 patients (7 eyes) in Group A experienced visual distortion. After 3 months of follow-up, all 7 patients had relieved visual distortion. Group B patients maintained stable retinal membrane structure during the follow-up period. Conclusions: PSR is a safe and effective method for pathological myopia. In short-term follow-up, the effect of shortening the axial length is better in adults than in children. LHMM24003 DIAGNOSTIC VALUE OF OAS3 IN CHILDHOOD-ONESET SYSTEMIC LUPUS ERYTHEMATOSUS Yangfan Huanga,b,c, Qinchun Pand, Zhijian Zhongb,c, Junming Wua,b,c aDepartment of Basic Science, YuanDong International Academy of Life Sciences, Hong Kong 999077, China,bExperimental Center of BIOQGene, YuanDong International Academy of Life Sciences, Hong Kong 999077, China,cSystems Biology Research Center, Biology Institute, Guangxi Academy of Sciences, Nanning 530007, Guangxi, China,dSchool of Medicine & Health, Guangxi Vocational & Technical Institute of Industry. Background: Childhood onset systemic lupus erythematosus (cSLE) is a complex multisystem autoimmune disease, characterized by immune dysregulation and systemic inflammation, leading to progressive and irreversible multi organ damage. The objective of this study is to identify diagnostic markers for cSLE based on bioinformatics analysis and machine learning algorithms. Subjects and Methods: Gene expression profiles for cSLE were obtained from GPL96 and GPL97 platforms of GSE8650, and GSE11909 datasets then merged de batches. Differentially expressed genes (DEGs) were identified in both platforms and common DEGs with consistent direction of expression were identified through intersection analysis. Then we performed enrichment analysis for common DEGs. Common DEGs with area under receiver operating characteristic curve (AUC) values > 0.8 in both GPL96 and GPL97 platforms were used to build risk model with XGBoost algorithm model. The most important variable in model was further identified as key gene. Possible immune regulatory mechanisms of key gene were explored by identifying association with immune cells, which analyzed by single-sample gene set enrichment analysis, and cell death that differed between cSLE and control were also analyzed. Results: A total of 4219 DEGs in the GPL96 platform, and 4860 DEGs in the GPL97 platform, with 314 DEGs were upregulated expression with the same direction in GPL96 and GPL97, and 314 DEGs were downregulated expression with the same direction. Then 628 common DEGs were identified, they enriched in immune response, neutrophil extracellular trap formation, and NOD−like receptor signaling pathway. There were 26 common DEGs with AUC values > 0.8 in both GPL96 and GPL97 platform and then a XGBoost model was constructed. Subsequently, OAS3 was identified as a key gene with the biggest AUC value and important ranking. In addition, activated DCs were higher in cSLE and positively correlated with OAS3. Apoptosis, autophagy, necroptosis, and pyroptosis were significantly activated in cSLE and positively correlated with OAS3. Conclusion: This study constructed a diagnostic model and identified OAS3 as a key diagnostic marker. OAS3 may regulate immune responses and NOD−like receptor signaling pathway involved in cSLE.
