Abstract: Amyloidosis is a disorder of protein metabolism in which characteristic abnormal extra cellular protein material is deposited in organs and tissues. Amyloid deposits may be focal, localized or systemic in distribution and are composed largely of protein fibrils, the nonglycosylated peptide subunits which are of different chemical constitution in different forms of the disease. Amyloid plaques are composed of a protein fragment called Beta amyloid, which is a string of 40 – 50 amino acids snipped from a larger protein called Amyloid Precursor Protein or APP. Beta amyloid forms tiny channels in neuron membranes, which allow uncontrolled amounts of calcium into the neurotic, an event lethal in any cell. It also disrupts potassium channels, which could also affect calcium levels. The self assembly of β sheet domains resulting in the formation of pathogenic fibrillar protein aggregates (amyloids) is a characteristic feature of various medical disorders. The present investigation was carried out to study the mechanism underlying amyloidosis and similar processes occurring in neurodegenerative diseases such as Alzheimer’s disease, Huntington’s Disease, Creutzfeldt Jacob’s Disease. A synthetic model peptide poly (Ile – glu) was chosen for our study, to evaluate its toxicity in lymphocytes. Peripheral blood lymphocytes were isolated from the healthy donor the isolated lymphocytes were exposed to the synthetic peptide and incubated for a week. After the incubation period cells were harvested and the toxicity studies such as cell viability assay, Apoptosis, Oxidative damage, Antioxidative enzymes. The Objective of this project included Synthetic model peptide poly (Ile – glu) was chosen for our study to evaluate its toxicity,Peripheral blood lymphocytes was isolated from the healthy donor,Isolated lymphocytes exposed to the peptide and incubated for a week. After incubation period cells were harvested and the toxicity studies were performed
