Effects Of Short-term Tai Chi On Circulating Oxylipins And Endocannabinoids In Post-menopausal Women

Year: 2024
Type: article
Abstract: BACKGROUND/OBJECTIVES: We recently reported differences in circulating levels of pro-inflammatory oxylipins (OxL) and endocannabinoids (eCB) among postmenopausal women with knee osteoarthritis after 8 weeks of Tai Chi (TC) (Shen et al. 2023, doi: 10.3389/fmed.2023.1210170). This study examined if plasma eCB/OxL levels change after 4 sessions of TC within a 10-day period in postmenopausal women. METHODS: Sixteen post-menopausal women completed 4 TC sessions on non-consecutive days within 10 days, 60 min/session including 5 min of warmup, 50 min practicing TC, and 5 min of cooldown. Participants learned 10 selected moves from 24-form Yang-style TC in sections designed to allow self-practicing in a short time. Plasma isolated from non-fasting blood draws at baseline and immediately after TC session 4 were analyzed for eCB (e.g., AEA and 2-AG) and OxL using targeted metabolomic methods that preserve 2-AG (Pedersen et al. 2021, doi: 10.1016/j.aca.2020.11.019). Specifically, oxylipins were isolated by solid phase extraction on 10 mg Oasis-HLB sorbent beds (Waters Corp, Milford, MA) as previously described (Luria et al. 2007, doi: 10.1074/jbc.M608057200), modified for use in a 96 well plate format. Partial least squares discriminant analysis highlighted changes between pre- and post-intervention samples. Paired t-tests with false discovery rate corrections were performed to examine the differences in plasma OxL and eCB. RESULTS: 16 women (58.9 ± 5.2 years old, BMI: 33.6 ± 5.4 kg/m2) completed the study. Few fatty acid values differed from baseline after the 4th TC session. The plasma 20:5n3 metabolite 18 HEPE, 18:2n6, 18:3n3, the 18:3n3 OxL metabolite 15,16-DiHODE, and multiple 18:2n6-derived metabolites were increased, while monoacylglycerols (e.g., 1-AG) and the 12-lipoxygenase metabolites 12-HETE and 14-HDoHE were lower after TC. CONCLUSIONS: The short-term 4 TC sessions in post-menopausal women had relatively little impact on eCB and OxL in self-reported healthy post-menopausal women. The lower levels of monoacylglycerols and higher PUFA 18:2n6 suggest that TC might mobilize fatty acids and involve enzymes of 1-AG metabolism such as phospholipase C, DAGL-alpha, MGL, and perhaps FAAH. A limitation in this study is the non-fasted state of the subjects.
