Abstract: • [ 18 F]AlF-HBED–CC–FAPIs can be readily prepared via a one-pot fluorination reaction. • [ 18 F]AlF-HBED–CC–FAPIs show excellent stabilities in ethanol or saline. • [ 18 F]AlF-HBED–CC–FAPIs display high FAP-specific uptakes in FAP-positive cells. • [ 18 F]AlF- 6 exhibits excellent and prolonged tumor uptake in tumor-bearing mice. • [ 18 F]AlF- 6 is a promising 18 F-FAPI agent suitable for clinical applications.
