Analysis of chicken LGALSL (galectin-related protein) gene’s proximal promoter and its control by Krüppel-like factors 3 and 7

Abstract: Galectins are proteins that recognize and bind β-galactosides by a conserved canonical sequence signature in the carbohydrate recognition site. They are pivotal in widespread processes like cell communication, immunoregulation, inflammation, and cancer development. The Galectin-Related Protein (GRP), encoded by the LGALSL gene, is a non-canonical member of this protein family with lost carbohydrate-binding abilities and has supposedly oncogenic potential in several human cancer types. Its chicken version (C-GRP) occurs in the bursa of Fabricius’ epithelial and B cells. This study aims to characterize the 5' region (-2480 bp and +26 bp) upstream of the chicken (C-)LGALSL gene and to identify potential transcription factors regulating promoter activity. Deletion variants of the sequence stretch from -2480 bp to +26 narrowed down the proximal promoter activity to the area of -274/+26 by luciferase reporter assay. There is evidence that Krüppel-like factors (KLFs) 3 and 7 can control the promoter activity of the -274/-75 promoter sequence of the C-LGALSL gene in a balanced way. Since there is no evidence for GRP’s functional role, the connection to KLFs could help find GRP-related pathways.
