Abstract: Introduction: Persons with type 2 diabetes mellitus (T2DM) show a notable increase in oxidative damage to DNA, which can be evaluated with 8-hydroxy 2-deoxy-guanosine (8-OHdG). The present study intended to evaluate the serum concentrations of 8-OHdG and lipid profiles in individuals with and without T2DM and to investigate any potential correlations between clinical traits and biomarker concentrations. Materials and methods: The current study involved 135 participants comprising 45 T2DM patients and 90 healthy controls and was done at a tertiary healthcare facility in South India. The venous blood samples were collected and analyzed. The plasma was subjected to a sandwich enzyme-linked immunosorbent assay to quantify the concentrations of 8-OHdG. Statistical Package Social Sciences (SPSS) version 26.0 for Windows was used to analyze the recorded data. Results: The average age of the participants in the study was 49.3 ± 7.83 years. The diabetic group exhibited a substantial rise in BMI, FBG, HbA1c, and lipid markers compared to the control group (p<0.0001). The level of 8-OHdG (905.79 ± 183.85 pg/ml) was significantly higher (p<0.0001) in the T2DM group compared to the control group (258.13 ± 117.27 pg/ml). The odds ratio of BMI and HbA1c that were stratified were combined to determine the Mantel-Haenszel adjusted odds ratio of 3.34, indicating an approximately three-fold increased risk of developing T2DM in individuals exhibiting increased levels of both BMI and HbA1c. Conclusion: The study found that 8-OHdG, LDL, BMI, and HbA1c can be used as predictor variables to determine the probability of developing diabesity.
