Type: article
Abstract: Electrospinning is a prominent technique in the field of drug delivery system (DDS) for manufacturing matrices. To achieve prolonged retention and controlled release of the drug at the lesion site, electrospun chip was fabricated and evaluated. A biodegradable electrospun chip loaded with chlorhexidine (BESC-CHX) was manufactured for the treatment of periodontal disease. Then, the physicochemical properties and efficacy of BESC-CHX were investigated. CHX content in BESC was analyzed using high performance liquid chromatography (HPLC). The antimicrobial activity and anti-inflammation effects of BESC-CHX were assessed using a disc diffusion assay and a periodontal disease model. BESC-CHX composed of 700 nm fibers was observed using scanning electron microscope (SEM) analysis. Many batches of BESC-CHX maintained a consistent levels of CHX. It released CHX for 7 days and was completely degraded in artificial saliva. BESC-CHX showed 99.99% of antimicrobial activity in 11 species of microorganisms. In the beagle periodontal disease model, BESC-CHX demonstrated improvements in periodontal probing depth (PPD) and clinical attachment level (CAL), but these changes were not statistically significant. However, alveolar bone regeneration was observed in cementoenamel junction-alveolar bone crest (CEJ-ABC) measurement. In addition, the lowest inflammation score was shown in this group. These results suggest that BESC-CHX could be beneficial and available for periodontal disease.
