Abstract: The aim of this present study is to formulate and evaluate the niosomal drug delivery system for Lamivudine utilizing non-ionic surfactant through Thin film hydration technique. The formulated niosomes were subjected to entrapment efficiency, scanning electron microscopy, in-vitro release, and Zeta potential analysis. The results indicates that formulation F7, which contains the drug at a surfactant to cholesterol ratio of 300:200 µmol, exhibited a higher percentage entrapment along a with desired sustained release profile of Lamivudine and thus formulation F7 was considered as optimized formulation.In-vitro release study of optimized Lamivudine niosomal formulation (F7) demonstrated an extended release up to 24 hours.Scanning electron microscopy images confirmed that the vesicles are spherical and uniform in size, with no observable aggregation among the particles. The Zeta potential analysis revealed a negative value, indicating the presence of a negative charge-inducing agent within the formulation.A stability study conducted over three months under various storage conditions indicated that the formulation remained stable at 4°C.The optimized formulation exhibited a zero-order release pattern, as evidenced by the linear relationship observed in the Time versus cumulative percentage drug release plot.Kinetic studies of drug release suggested that the drug was released from the niosome via a zero-order diffusion-controlled mechanism. This study illustrates that the formulated niosomes provide extended release for up to 24 hours while maintaining greater stability.
