Abstract: Introduction:The focus of the study was to evaluate the oral bioavailability of capsulated SuSEDDS of Rilpivine, a poorly bioavailable antiretroviral and to compare with an internal standard of Rilpivirine also administered as a capsule.Materials and Methods: Optimized F7 formulation of Rilpivirine for oral administration was developed.Stability studies were performed at 40±2°C/75±2% relative humidity for 3 months packed in Alu-Alu blister pack and HDPE container.Rabbits were used as the animal model for in vivo pharmacokinetic investigations Various key pharmacokinetic parameters like Tmax, Cmax, AUC, and t1/2 were identified and subjected to statistical analysis applying unpaired Student t-test.Results: A satisfactory stability profile was found for each formulation after three months of research, according to the stability study. The in vivo study of the optimised formulation demonstrated superior pharmacokinetic performance when compared with the internal standard of the drug. The Rilpivirine F7formulation exhibited a relative bioavailability improvement of around 2.57 folds. Conclusion:The in vivo performance of the therapeutic candidate may be enhanced by the development of Rilpivirine into Solid SuSEDDS as capsule dosage form.
