Abstract: This literature review provides current data on the oncosuppressive effect of exogenous nucleases derived from various sources. Exogenous nucleases, such as RNase A, BS-RNase, onconase, and DNase I, are capable of degrading low-molecular extracellular DNA and RNA circulating in blood plasma, which may significantly reduce the risk of metastasis. The role of extracellular DNA and RNA in oncogenesis, as well as the impact of exogenous nucleases on their levels, have been described. The review explores the prospects of using nucleases in combination with other therapeutic methods, such as chemotherapy and radiation therapy, to enhance their effectiveness and reduce side effects. In addition, the targets and mechanisms of action of nucleases, as well as their potential for combined use with other therapeutic agents, are considered. The review concludes that exogenous nucleases have significant potential in the treatment of cancer.
