Abstract: This study explores the formulation and characterization of Neomycin sulfate-loaded nanosponges (NS) to enhance the skin penetration. Utilizing the emulsion solvent evaporation technique, we developed NS encapsulating Neomycin sulfate, which is a BCS Class III drug known for its poor skin permeability. Various formulations were optimized using Box-Behnken design, with independent variables including the concentration of ethyl cellulose and polyvinyl alcohol, and the stirring rate. Characterization of the NS indicated a high entrapment efficiency of 89.6%, an average particle size of 236.1 nm, and a zeta potential of -13.6 mV, confirming their stability. The resulting NS gel exhibited favorable physical properties, including a neutral pH and good spreadability. In vitro drug release studies revealed a controlled release profile, with the Korsmeyer-Peppas model being the most applicable for release kinetics. Stability studies confirmed the formulation's integrity over a six-month period. These findings suggest that NS can effectively enhance the delivery of Neomycin sulfate, potentially improving treatment outcomes for skin infections.
