Immunodiagnosis of Autoimmune Myopathies

Year: 2024
Type: other
Abstract: Autoantibodies in Autoimmune Myopathies (AIM) have been tested by a combination of immunoprecipitation (IP) analysis of proteins and RNA components of the target antigens along with enzyme-linked immunosorbent assays (ELISAs) using purified or recombinant antigens, Western blotting, and IP-Western blotting as necessary. This chapter focuses on experimental procedures of IP and ELISA using recombinant proteins. As a basic rule in interpretation of IP, observation of any bands that are present in a particular lane but not in other lanes means that they represent proteins specifically recognized by autoantibodies in the respective serum. The quality and reliability of ELISA depend heavily on the quality of the recombinant protein used. Immunoprecipitation is a powerful technique and is considered the gold standard for many myositis-specific autoantibodies; however, it is time-consuming and labor-intensive, in addition to its use of radioisotopes.
