Abstract: Background: The study had performed mainly to investigate toxicopathological changes in rat pups exposed to low toxic dose of lead acetate through dam’s milk and the protecting role of Alpha lipoic acid and, or calcium gluconate with induced lead toxicosis. Materials and Method: Seventy-five adult albino rat consist of 25 male and 50 female divided equally into five groups, four dosed daily orally with lead acetate at a dose 20mg/kg B.W represented control positive (C+ve). Other treatment groups with drugs at same dosing regimen with calcium gluconate 500mg/kg B.W represent (T1-Ca), ALA, 60mg /kg B.W (T2-ALA) and their combination at half dose(T3-Comb) and distilled water for control negative group (C-ve). The pups euthanized after lactation period and the MDA and Gpx estimated from serum. Brain, liver and kidney submitted to histopathological sections. The statistical analysis don by using SPSS V.20, one way ANOVA at significant (0.05) ≥ p- value. Results: The results of MDA and Gpx enzyme revealed that C+ve group had the most significant elevations for MDA followed by T2,T1, T3 and C-ve group while the lest significant decline for Gpx enzyme followed by other treated group at same arrangement. Gross toxicopathological changes for C+ve group showed grossly brain tissue with hemorrhage under skull, congested blood vessels while some vessels appeared filled with blood embolism in cerebrum and cerebellum. The histopathological section in brain of C+ve group showed congested blood vessels in choroid plexus, proliferation of glial cells with necrosis of neuron that contact with oligodentritic cells, congested parenchyma with less lesion were seen in T2 followed by T1 group, while T3 combined group showed nearly normal brain section as C-ve group. The liver and kidney of C+ve treated rat pups group, showed many lesions, degreed from mild to severe lesions. While improvement were recorded for T2 followed by T1 and T3 groups that showed less degenerative effect in both liver and kidney section. We concluded that lead acetate poisoning in rats causes toxicopathologiacl changes in the examined organs lesions. Conclusion: Our conclusion had been referred to that increase in oxidative stress may be the main cause to the great damage on pups brain that have been caused by lead toxicity. While the supplementation with Ca, ALA and their combination act to improve and alleviated lead toxicity especially in combined treatment group.
