Association between physical activity and prostate cancer: A Mendelian randomization study

Abstract: Objectives Prior observational research has revealed a substantial correlation between insufficient physical activity (PA) and increased vulnerability to prostate cancer (PCa). To further explore this correlation, we performed a two-sample Mendelian randomization (MR) analysis using genetically predicted PA and PCa data. Methods Our investigation used substantial PA and PCa datasets comprising 377,000 and 182,625 individuals, respectively. Causal effects were estimated using various methods, including simple mode, MR-PRESSO, weighted median, weighted mode, and inverse-variance weighted (IVW) analysis. Results The outcomes of our investigation suggest that a higher genetically predicted accelerometer-assessed average acceleration PA is associated with a reduced risk of PCa (OR per SD: 0.995; 95% CI: 0.993–0.998, P < 0.01 for the IVW analysis). Nevertheless, no causal relationships were observed between vigorous physical activity, moderate to vigorous physical activity, the fraction of accelerations exceeding 425 milligravities in PA, and a reduced risk of PCa. Conclusion Higher levels of PA may mitigate the incidence of PCa. Therefore, promoting active lifestyles on a broader scale could alleviate the burden of the most prevalent cancer among men.
