Abstract: Supplementary Figure S15. TR-107 increases cellular and mitochondrial ROS, leading to mitochondrial membrane permeabilization and lipid peroxidation. MITOSOX Red levels were measured in HCT116 (A) and DLD-1 cells (B) over a three-day period. Doxorubicin (DOX) was used as a positive control. Cellular ROS levels were assessed with DCFDA in HCT116 (C) and DLD-1 cells (D) over a three-day period with doxorubicin (DOX) as a positive control. The mitochondrial membrane permeability was measured using the TMRE assay kit and FCCP as a positive control in HCT116 (E) and DLD-1 cells (F) at 24, 48, and 72 hours. Lipid peroxidation assay was determined using MDA assay using doxorubicin (DOX) as a positive control in HCT116 (G) and DLD-1 cells (H) at 24, 48, and 72 hours. Data are presented as mean ± SD values (N=3). *, P < 0.05; **, P < 0.01; ***, P < 0.001; ****, P < 0.0001.
